Production and Characteristics of Foreign Glucoamylase from Recombinant Saccharomyces cerevisiae

|
The optimal medium components and composition for foreign glucoamylase production and the characteristics of cloned foreign glucoamylase were investigated for a recombinant yeast system. This system consisted of the yeast Saccharomyces cerevisiae transformed with the 2 ¥ì-based recombinant plasmid YEpSUCSTA which contains the SUC2 promoter and glucoamylase coding STA gene including the signal sequence and structural gene. Large amounts of foreign glucoamylase were produced in a medium containing glucose and starch. It was thought that the recombinant yeast MMY2SUCSTA utilized starch as a carbon source for glucoamylase biosynthesis. The optimal concentrations of glucose and starch were both 10 g/L. Yeast extract was an important factor for foreign glucoamylase biosynthesis while the optimal concentration was 10 g/L. The optimal pH and temperature for the activity of purified glucoamylase were 5.0 and 60¡É, respectively. Kinetic parameters V_(max) and K_m involved in the Michaelis-Menten equation were calculated as 0.509 g/L¡¤min and 2.28 g/L, respectively.
